Synthesis and Biological Evaluation of Some Novel pyrido[2,3-b]pyrazine derivatives as Potent TNF?alpha? inhibitors

Two series of 3-Methyl-pyrido[2,3-b]pyrazine-2-yl)substitutedphenylamines 3a-f and N-(3-methylpyrido[2,3-b]pyrazin-2-yl)substituted benzenesulfonamides 4a-f were synthesized and evaluated as novel TNF-? inhibitors. The latter compounds were tested for their ability to inhibit the production of TNF-? in vivo in rats. Compounds 4b, c and d exploited potent inhibitory activity with IC 50 = 0.025, 0.040 and 0.053 nM respectively.Downloads
